Hi
From the error message, it seems that you didn’t define the managed path for the web application which holds “mysite” site collection. Please go to “Define Managed Path” to check it according to the Chris Johnson’s blog provided by Serge Luca. I suggest that you create an explicit inclusion for path “mysite” and a wildcard inclusion for path “personal” as Johnson said. When you create the site collection for “mysite”, make sure the URL of site collection look like http://moss:1000/mysite. As a result, “your “personal site provider” should be the site collection address like: http://moss:1000/mysite and the location is “personal” in “My Site Settings”.
Hope this helps.

- MySite questions go to Social Computing.Moving thisLater:NEVER NEVER post the same question to two forums. This was originally posted to General, yet after I moved it to Social Computing you also posted a copy of it to Setup/Admin. This wastes everyone's time. Don't please do it again.
