This means that the remote host (web.tnmgrmu.ac.in) is not responding or not responding properly.
Instal TMG Best Practices Analyzer and use TMG Data Packager and run it in repro mode for web proxy and look at the resulting data when sending requests to this site. Focus on netmons and webproxy logs. Do note the IP address of the client making the request in order to find the request in question easier.
Run the repro for as short time as possible as it may generate a LOT of data...

Please check if there is any other firewall in front of the ISA server or if there is the upstream firewall, it is usually the one dropping the connection.
