MOSS 07 Permission: Department permission restriction for certain Site collection

Our firm is coming up with a new department that need to be restricted from accessing certain site collection. We are building a new site collection for the new department.
What I am looking for is if someone mistakenly gives a member of the restricted department permission to a restricted site, the person should not be able to access the site irrespective of the access level.
Please let me how to achieve the above problem.

if this was a different web application you could set a 'deny policy' easily.

No, not related.
As BlueSky2010 proposed, you can create a new web application and then use the Deny option at the application level. This would not require a new web front end (WFE), but would require an additional URL. (example: newdepartment.contoso.com instead of www.contoso.com/sites/newdepartment)
