    I would like to implement 1 WSUS server and create Target groups for my 3 domains Test, Acceptance and Production.

    The domains Test, Acceptance and Production are NOT trusted.

    Will client-side targeting work here by using GPO's ?

    It will work. Configure your firewalls, and point the clients to the WSUS server. You may need to point the clients to the IP address of your WSUS server if it can't be resolved by name.

    And client-side targeting will work as well ?

    So when I create at the 3 domain's GPO's with the different Target group set then the will be added to the correct group at WSUS ?

    The client-side will work as well, just make sure in WSUS you configure Use Group Policy or registry settings on computers from Options > Computers.

    A point of clarification, which may help here.

    The WSUS server does not care how the WUAgent gets it's configuration, whether it's by GPO, LPO, or somebody hacking the registry. As such the WSUS server also does not care about the client system's domain membership, any trusts that may or may not exist, or the fact that the client is not a domain member at all.

    The client connects to the WSUS server anonymously, so all that matters is that the client knows where to connect.
