Hi Yasser,
Does the client fail to update? If yes, which error code is shown in the update?
If it is possible, please reproduce the issue then post the entire windowsupdate.log of the client here.
Beside, as Lawrence has mentioned above, have you appended the ":8530" to the FQDN of the WSUS server?
We can verify this by checking the registry below,
HKEY_LOCAL_MACHINE\SOFTWARE\Policies\Microsoft\Windows\WindowsUpdate
Here is the screenshot of my lab server,
Best Regards.

Trying for a client to download this file http: // {name-of-the-wsus-server} /iuident.cab (replaces {name-of-the-wsus-server} with the name of your server wsus
Finally you can send me your stp file c: \ windows \ WindowsUpdate.log

You have in your file this error :
Failed to get Wu Exemption info from NLM, assuming not exempt, error = 0x80240037
Can you try this:

Execute the command line (on the wsus server) with admin right
wsusutil usecustomwebsite true;
This command change the port on IIS to use 8530 port. It's necessary to change the value in GPO

We upgrade our wsus from 3.0 to wsus 2012 r2
Lots of replies here... let's go back to the beginning.
This is a functionally impossible scenario.... so what did you REALLY do?
Note: If you upgraded an *OS* (e.g. WS2003/WS2008/WS2008R2) to WS2012R2, then the short answer is: It's TERMINALLY broken, and this scenario is explicitly documented.
Answer: Start From Scratch.

I think your problem is selfupdate folder. Can you please test the command below :
Can you try execute the command dos wsusutil /checkhealth and look into the event Applications. There is normally one event for wsus or more (if wsus don't work)
