we need to know whether wsus server license is required for window server 2012 r2 std edition.

Hi,
WSUS is a feature of Windows Server. So yes, if you wish to run a WSUS instance, you have to properly license the Windows Server instance it will be running on. Apart from that, no further server licensing is required.
In a virtualized environment that should have been already taken care of by host licensing.
Of course you also need to properly license the device access to that WSUS instance. For every device you point to it for updates there has to be a valid CAL assigned to either the device itself (device CAL) or its primary user (user CAL).

Thanks for the information.
We are planning for WSUS Server in Virtualization (VMware) and we have SQL Server Enterprise License also for the database.
is there any other license is required for WSUS.
We have nearly about 300 computer including 20 Servers.

if you have Windows Server licenses to support another server instance and CALs to support the existing client access, then no, you're all set.
