Hi noudre,
There are two level Out-going Email setting for WSS 3.0. It can be configured in the farm in the following path:
Central Administration > Operations > Outgoing E-mail Settings;
It also can be configured for particular web application:
Central Administration > Application Management > Web Application outgoing E-mail Settings
So please check whether it was configured correctly for the web application.

Hi,
Check these :
1. Check if all IPs of your WFE servers are added into the allow relay list of your SMTP server.
2. Check the web application email settings as described by Wayne.
I hope this will help you out
Thanks, Rahul Rashu
