Depending on the infrastructure model you are going to use, you might not even need that one server, see: http://technet.microsoft.com/en-us/library/cc843778.aspx for Planning and Deployment Guide for the Application Virtualization System. There's also App-V 4.5 Infrastructure Planning and Design Guide at http://technet.microsoft.com/en-us/appvirtualization/cc843994.aspx which should get you forward.

Also, the App-V Trial Guide whitepaper was very useful to me. It provides step-by-step instructions to set up your own App-V trial environment. I would recommend you give that a shot if you're just starting out.
