Nope. The primary auth is either FBA, WIA or Cert (FBA and cert on a WAP) for Windows Server 2012 ADFS.
For ADFS 2016, this list has been extended to Windows Hello for Business as well as Azure MFA (as a primary factor).
There is no possibilities to add your own.
