No. Setup constrained delegation and you can move the virtual machines to your 2012 R2 server without downtime.

Microsoft has never supported operating system upgrades within a cluster.
http://blogs.msdn.com/b/clustering/archive/2012/06/25/10323434.aspx explains the process to move from 2008 to 2012, but the same process will work to move from 2012 to 2012 R2.
