As Anoop suggested you should deploy a stand-alone Primary Site in China (with two management points). I would also consider using secondary sites in all the other locations.

According to your statement 1 primary in china and two secondary in India and one secondary in Singapore will do.
Yes, I think so. However I don't know whether MP rotation (when you've more than one MP) will be a problem for you as in my experience it's bit of challenge if your clients are in same domain or forest.
Some More details about MP rotation this post is about DMZ or untrusted forest however this will be more complex issue when all your client is in same domain http://anoopcnair.com/2014/04/11/workaround-sccm-2012-clients-mp-selection-rotation-issue-untrusted-dmz-forests/

No need to have CAS ! Head office with Primary. However I can't tell you whether you need to have secondary servers in other remote offices as this needs to be analysed in bit details as mentioned in the below paragraph.
We can't provide the details of design via forums as that involves loads factors like connectivity between the locations, all the clients are in same domain or forest ,which are the functionalities you're going to use , Hardware Inventory, Software Inventory, Software Updates and so on...
