  Check queue on servers and check send connector settings.

  Did you make any changes to the default connectors?

  Try to review the connector protocol logs for further analysis.

  Thanks for contacting our forum.

  From your description, the migrated mailbox to 2016 cannot send messages, what about the mailbox on Exchange server 2010? If they can, please compare the properties of the mailboxes between the migrated and didn't migrate.

  Firstly, please check the queue with the command get-queue | fl to check if any error message in Last Error part, please post out in detail if exist.

  It's recommended to restart the Transport Service on all of your servers and check the result.

  Plase also post out the receiveconnector and sendconnectors' information of Exchange server 2016 via the command below:

  • Get-sendconnector | fl
    
  • Get-receiveconnector | fl
